چکیده

Recent observations on conjugated proteins (l-6) raise the question as to whether the prevailing ideas concerning the nature of the attachment of nucleic acids with the proteins are valid. With certain exceptions it has generally been assumed that the nucleoproteins are salts of a polyvalent nucleic acid and a polyvalent base (protein) (7). The ideas that nucleoproteins are salt-like (polar), or dissociable combinations, are largely derived from results of experiments in which vigorcpus chemical methods have been used. In general the following methods have been employed: (a) extraction of cells and organs by a dilute alkaline solution, and the precipitation of the nucleoproteins with acid (8) ; (b) treatment of the starting material with acetone, or alcohol and ether; the dry material then extracted with water, saline, buffer, or alkali, and the nucleoprotein precipitated with acid (9, 10); (c) extraction of the acetoneand ether-dried material with 10 per cent saline at 70-80” for several days and the nucleoprotein precipitated with acid (11). The idea of “salt-like” combination has received further emphasis from the fact that the nucleoproteins prepared as above, and the artificial “nucleoproteins” (protein nucleates) precipitated from a mixture of nucleic acids and proteins, at times have comparable analytical results and show similar properties (7, 12, 13). This misconception is easily explainable if we consider that many so called nucleoprotein preparations are protein nucleates, or are mixtures of nucleic acids and proteins due to the splitting of the

Skin Reactions to Nucleoprotein of Streptococcus Scarlatinae in Patients with Rheumatoid Arthritis and Rheumatic Fever.

The skin reactions to various products of the hemolytic streptococci have been studied by a number of observers, and attempts have been made to correlate the results of such tests with various clinical manifestations of streptococcal infection. In the present investigation the skin reactions to varying amounts of nucleoprotein derived from a strain of Streptococcus scarlatinae were studied in 2...

Prokaryotic Expression of Influenza A virus Nucleoprotein Fused to Mycobacterial Heat Shock Protein70

Background and Aims: The novel approaches in influenza vaccination have targeted more conserved viral proteins such as nucleoprotein (NP) to provide cross protection against all serotypes of influenza A viruses. Influenza specific cytotoxic T lymphocytes (CTL) are able to lyse influenza-infected cells by recognition of NP, the major target molecule in virus for CTL responses. On the other hand,...

Evaluating the Immunogenicity of Avian Influenza Virus Nucleoprotein

Background:  Influenza viruses cause Avian Influenza (AI) is a serious infectious disease belonging to type A Orthomyxovirus. A viral RNA synthesis is due to an interaction of the nucleoprotein (NP) with the viral polymerase. In the present study, we have evaluated the immunogenicity of avian influenza virus nucleoprotein.  Materials & Methods:   An Influenza Virus N9H2 subtype A/Chicken I...

Optimization of Lysozyme – Inulin Conjugation and Investigation on its Functional Properties

In recent years, several oconjugation protocols have been developed to improve the functional properties of the enzyme lysozyme. In this study, the optimum conditions of lysozyme – oxidized inulin conjugation and functional properties of the modified lysozyme were investigated. Initially, inulin (MW about 25 kDa) was oxidized by periodate to provide the reactive carbonyl groups  to attach to am...

Studies in the Relation of the Hemolytic Streptococcus to Rheumatic Fever

AFTER DISINTEGRATION BY SONIC VIBRATIONS THE CONTENTS OF THE HEMOLYTIC STREPTOCOCCUS CAN BE SEPARATED BY DIFFERENTIAL CENTRIFUGATION INTO THREE FRACTIONS: an insoluble residue, cytoplasmic particles, and a solution of proteins of smaller unit size. The residue (R) presumably comprises the cell walls of the bacteria and contains the type-specific M protein. The cytoplasmic particles (CP) contain...
